Research Interests

The morphology of cells, organs and whole organisms is determined by the generation of forces on the immediate environment, which is either extracellular matrix or adjacent cells. We are currently engaged in studies to understand the detailed molecular mechanisms involved in a variety of phenomena from cancer metastasis to brain function. Further, we are developing several new tools and protocols for measuring cell forces at the molecular level, which are revealing many new aspects of how cells can both generate and respond to external forces. We have an effort underway to define quantitatively the steps involved in cell adhesion to and spreading on a matrix-coated surface. Using a variety of cell lines that are missing proteins in various motility pathways, we are determining the quantitative changes in the spreading process. This will enable us to generate a detailed model of the process of spreading that will be a model for further studies of how cells differentiate, regenerate tissues or metastasize.

Biography

Hailing from Columbia University, Prof Michael Sheetz has more than 40 years’ experience in the biomedical field. Introduced to Singapore by Prof Hew Choy Leong of the National University of Singapore’s Department of Biological Sciences, Prof Sheetz was sought to lead an RCE project. This resulted in a two-year effort to organise and submit a proposal on Mechanobiology where Prof Sheetz set the theme and direction of the Mechanobiology Institute (MBI). As Founding Director of the MBI, he led the institute for 10 years and built it into a world-leading research centre in the field of mechanobiology.

What are chromatin, heterochromatin and euchromatin?

Nov 30th, 2023|Comments Off on What are chromatin, heterochromatin and euchromatin?

What is chromatin, heterochromatin and euchromatin? The human genome contains over 3 billion base pairs or nucleotides. These nucleotides, which are arranged in a linear sequence along DNA (deoxyribonucleic acid), encode every protein and [...]

What are chromosomes and chromosome territories?

Nov 30th, 2023|Comments Off on What are chromosomes and chromosome territories?

What are chromosomes and chromosome territories? While metaphase chromosomes can be depicted as distinct bodies with well-defined shapes and sizes, interphase chromosomes are less uniform and, by filling the nuclear space, difficult to distinguish. [...]

How does chromatin condensation keep the nucleus compact?

Nov 30th, 2023|Comments Off on How does chromatin condensation keep the nucleus compact?

How does chromatin condensation keep the nucleus compact? Packaging of approximately a meter-long DNA duplex within the elastic nucleus determines its spatial 3D architecture and the genomic accessibility of tissue-specific transcriptional programs. Apart from [...]

How do chromosome territory dynamics affect gene redistribution?

Nov 30th, 2023|Comments Off on How do chromosome territory dynamics affect gene redistribution?

How do chromosome territory dynamics affect gene redistribution? The spatial organization of chromatin within the 3-dimensional space of a chromosome territory enables the co-localization of co-transcribed genes and their transcriptional foci. Many gene positioning [...]

How does chromatin remodeling impact gene transcription?

Nov 30th, 2023|Comments Off on How does chromatin remodeling impact gene transcription?

How does chromatin remodeling impact gene transcription? While chromosome territory dynamics is believed to regulate gene expression through the redistribution of genes and the subsequent co-localization of these genes with transcription machinery, changes are [...]

How is transcription regulated in stem cells?

Nov 30th, 2023|Comments Off on How is transcription regulated in stem cells?

How is transcription regulated in stem cells? Pluripotent stem cells can potentially differentiate into any given cell type. They commonly exhibit mechanically softer nuclei [1], lack type-A lamins [2] and have a poorly defined [...]

How is DNA replicated?

Nov 30th, 2023|Comments Off on How is DNA replicated?

How is DNA replicated? Cells must replicate their DNA before they can divide. This ensures that each daughter cell gets a copy of the genome, and therefore, successful inheritance of genetic traits. DNA replication [...]

What is translation?

Nov 30th, 2023|Comments Off on What is translation?

What is translation? Translation is a process that involves the synthesis of an amino acid chain from an mRNA blueprint. These polypeptide chains fold into functional proteins. Translation occurs outside the nucleus once nuclear [...]

What are transcription factories?

Nov 30th, 2023|Comments Off on What are transcription factories?

What are transcription factories? The primary role of chromatin assembly is the compaction of an otherwise very long DNA molecule into discrete structures that fit into the nucleus. However, this compaction cannot be achieved [...]

How does the heart form?

Nov 30th, 2023|Comments Off on How does the heart form?

How does the heart form? The heart is derived from the splanchnic lateral mesoderm and initially forms as two crescent-shaped endocardial plates inside the evolving pericardial cavity. As the embryo undergoes lateral and cranial [...]
