SUMMARY:Huygens Deconvolution\, Visualization and Analysis Workshop by MBI Microscopy Core
DESCRIPTION:MBI Microscopy Core will be holding Huygens Deconvolution\, Visualization and Analysis Workshop at MBI\, level 5 seminar room on 2nd October 2017. The workshop will start with an entry level class from 9am to 12pm\, followed by an advanced level class from 12.30pm to 5.30pm. You are welcome to register for both sessions to learn all the analysis tools in Huygens software or only register for one of the sessions. For registration\, please email Hui Ting (mbioht@nus.edu.sg) with the following details by 3rd September.  After the closing date\, successful registrants will be notified. \nSession 1 Entry level (9am-12pm)\nWe will start with a seminar on how to capture a good image for deconvolution and follow by hands-on training with Huygens deconvolution (Deconvolution Express\, Deconvolution wizard and the batch processor) \nSession 2 Advanced level (12.30pm – 5.30pm)\nThe advanced session will also begin with a seminar about Huygens deconvolution\, and follow by hands-on. Session 2 will be more interactive\, we will also focus on processing your own images with extra introduction with the Huygens Visualization\, Analysis tools and PSF distiller.

SUMMARY:Investigation of key steps involved in DNA Homologous Recombination Repair at single molecular level by Chen Jin
DESCRIPTION:Mechanobiology Institute\, Singapore (NUS)\nPhD Oral Defence \nInvestigation of key steps involved in DNA Homologous Recombination Repair at single molecular level \nSpeaker           :   CHEN Jin (Graduate Student\, MBI\, NUS)\nDate                 :   19 Oct 2017\, Thursday\nTime                :    10am\nVenue              :   T-Lab\, MBI\, level 5 seminar rooms\nSupervisor(s)  :   Prof YAN Jie \nAbstract: Homologous recombination (HR) repairs DNA double strand break through genetic exchange with identical chromosome under the regulation of a variety of repair proteins. Single-stranded DNA binding proteins such as prokaryotic SSB and its eukaryotic homolog RPA are important factors involved in HR. In this study\, activity of a group of E.coli SSB and human RPA on a single ssDNA molecule binding is investigated. In addition\, a label-free single molecule platform is developed to detect real-time annealing events between a mechanically extended ssDNA and short complementary ssDNA oligos. SSB and RPA showed different extent of inhibitory effect on annealing between ssDNA through retarding the nucleation process. Moreover\, a spontaneous homology specific interaction between ssDNA and dsDNA is observed\, indicating formation of a parallel DNA triplex structure. The parallel triplex has similar stability compared to B-form dsDNA. The triplex structure provides clue for the actual role of DNA recombinase in homologous search and pairing. In sum\, results in this study provide molecular insights for the key steps involved in HR. \nALL EXCEPT PANEL ARE EXCUSED FOR CLOSE DOOR EXAMINATION
