SUMMARY:MBIw: Development of the MBI Pillar Tracker Microscope by Felix Margadant
DESCRIPTION:MBI Weekly Meeting Seminar\nTime: 9.30am-10.30am\nDate: Friday\, 2 November 2018\nVenue: Level 5 Seminar Room\, T-Lab \nDevelopment of the MBI Pillar Tracker Microscope\nby Felix Margadant\, Senior Research Fellow\, Sheetz Group \nBrief summary\nThis talk summarizes the road of development and the status quo of a prototype light microscope dedicated to the fast or long term tracking of force deflecting pillars with time parallel recording of fluorescent images. \nThe machine is an evolution from the early far-red low numerical aperture condensers which we built in order to avoid photo toxicity by using transmitted light of 635nm or longer wavelength. This tracking proved to be fast (157fps) and repeatable to better than 4nm. Specimen distortions limit the universality of the approach though\, and the focal plane cannot be stabilized which triggered the design of the presented system. \nThe talk addresses the problems with back reflected light\, image distortion\, and speed limitations of the current system.
